Pigments from Algae

Materials and Methods

Sample Collection (Sept. 2019)
Live and healthy specimens were collected along the coast the Venetian Laguna, at the island La Certosa, and at the Adriatic sea coast of the Lido di Venezia, during low tides in the month of September 2019. The seaweeds were then washed thoroughly with seawater to remove extraneous materials and brought to the laboratory in plastic bags.

Sample Preparation
The samples were gently rinsed with fresh water to remove salt, sand, animal castings, calcareous and other adhering detritus matters. The samples were identified following the standard books. The seaweeds collected were identified as Ulva (green macro algae) and several Red Algae. The seaweeds were then cut into small pieces, weighed and subjected to pigment analysis.

Extraction Using Acetone
50 g of seaweed was kept in a pestle and mortar with 10 ml of acetone and it was ground well. The homogenate was macerated for 3 days in Acetone. Later it was centrifuged at 3000 rpm for 15 minutes and the supernatant was stored. The pellet was re-extracted by repeated washing with 5 ml of 80 % acetone till it became colourless. All the extracts were pooled and utilized for pigment quantification. The process was followed for all the samples.

Species

The cosmopolitan genus Ulva (Chlorophyta) Linnaeus includes species of green macroalgae found in marine, brackish and some freshwater environments. Although there is a wide literature for the determination of Ulva taxa in Europe, they are among the most problematic algae to accurately identify, because they have few distinctive features, as well as a high intraspecific variation.

Red algae, or Rhodophyta, are one of the oldest groups of eukaryotic algae. The Rhodophyta (red algae) are a distinct eukaryotic lineage characterized by the accessory photosynthetic pigments phycoerythrin, phycocyanin and allophycocyanins arranged in phycobilisomes, and the absence of flagella and centrioles.

Pigmenten van microalgen worden ingedeeld in de chlorofielen (groen), phycobili-eiwitten (rood, blauw, paars en geel) en carotenoïden (geel tot rood). De concentraties van deze pigmenten in de cel zijn vaak te laag voor commerciële exploitatie. Uitzonderingen zijn phycocyanine (blauw) in Arthrospira platensis (ook aangeduid met de oude naam Spirulina platensis), β-caroteen (oranje-rood) in Dunaliella salina en astaxantine (rood) in de alg Haematococcus pluvialis.
Phycocyanine is eigenlijk een complex van eiwitten en blauwe pigmentmoleculen. Het draagt in de cel bij aan het opvangen en transporteren van energie uit zonlicht. In Arthrospira platensis vormt phycocyanine ongeveer 15% van het totale drooggewicht.
Voor de meeste soorten microalgen zijn centrifuges nodig om de biomassa van het kweekmedium te scheiden.
Carotenoïden vormen één van de meest wijd verspreide groepen van natuurlijk voorkomende pigmenten. Ze komen voor in alle fotosynthetische en enkele niet-fotosynthetische organismen. De kleuren van deze pigmenten lopen van geel tot rood.
In fotosynthetische organismen helpen carotenoïden bij het vangen en transporteren van energie uit zonlicht. Ze geven structuur aan de ‘fotosystemen’ in de cel en zorgen voor eventuele afvoer van een overschot aan geabsorbeerde energie. Ook werken ze als antioxidant: ze ruimen reactieve zuurstofdeeltjes op. Voorbeelden van rendabele carotenoïdenproductie zijn op dit moment β-caroteenproductie met Dunaliella salina en astaxantineproductie met Haematococcus pluvialis. Haematococcus pluvialis is een zogenoemde mixotroof en wordt zowel gekweekt op organische koolstofbronnen als op zonlicht en CO2 in gesloten kweeksystemen. De carotenoïden worden als opgezuiverde stoffen en als componenten van gedroogde algenbiomassa verkocht.
Pigmenten zijn in relatief lage concentraties (minder dan 1% van het totale drooggewicht) aanwezig wanneer de cellen onder optimale groeicondities worden gekweekt. De microalgen gaan echter over tot ophoping van deze moleculen, tot wel 14% van het totale drooggewicht, wanneer ze worden blootgesteld aan suboptimale groeicondities, zoals een zeer hoge lichtintensiteit, een tekort aan stikstof of suboptimale temperaturen. De opgehoopte pigmenten functioneren vervolgens als een soort zonnescherm om de fotomachinerie tegen overbelichting te beschermen.
Omdat zowel β-caroteen als astaxantine pas worden opgehoopt onder omstandigheden die nadelig zijn voor de groei van de cellen, worden meestal ‘twee-staps’ productieprocessen toegepast. Eerst wordt een grote hoeveelheid biomassa geproduceerd onder optimale groeicondities. Vervolgens worden de microalgen aan stress blootgesteld om zo ophoping van de carotenoïden te stimuleren. In de praktijk betekent die stress meestal: stikstoftekort. In de groeifase van de algen is dat vrij eenvoudig te realiseren, doordat de groeiende algen de aanwezige stikstof zelf opmaken. Meestal wordt dit gecombineerd met verdunning van de cultuur, om zo de gemiddelde lichtintensiteit in de cultuur te verhogen.
