 +I followed the protocol we discussed,
 +- I took a nice piece of BC that I’ve cut in pieces of 20 grs each.
 +- I neutralised the BC as prescribed in the protocol, and I autoclaved the little pieces, together with a bit of BG11 still in the bottles.
 +- I’ve put the pieces in sterile petri dishes (working with the flame) and I inoculated them with Nostoc & Synechocystis (separately), on some petri's I added cleaned lichen on top of the cyanobacteria.
 +- Everytime I made 2 petri’s, to have a control. 
 +- I’ve put the petri's in the incubator at 22°C, on 25/2/2020.
 +- When I came back from Paris and when I controlled the petri’s, there was the same mould on all BC pieces.
 +This kind of ‘greenish blue cheese penicilline mould’. On the samples inoculated with Synechocystis, there was besides the blue/green mould also a kind of black mould.
 +- The samples with cyanobacteria + lichen had more mould than the ones without lichen. The samples inoculated with Synechocystis had more mould than the samples with Nostoc. The little black pieces on top of the BC on the petri’s with ‘lichen’ samples, are the lichen.
 +- The contaminated samles are not yet tested on their pH, neither I have inspected the (possible) growth and the mould under the microcope. This needs to be done asap.

 +Wat ik denk, betreffende die contaminatie, is het volgende:
 +- de Nostoc is gecontamineerd. De Nostoc die ik heb, heb ik reeds vanaf begin augustus 2019. Aangekocht bij Carolina.com voor ‘studiedoeleinden’. Geen gegarandeerd steriele samples dus. In de vloeibare culturen (die ook niet meer steriel zijn) zie ik helemaal geen contaminatie, maar in combinatie met de geneutraliseerde BC komt de contaminatie misschien naar buiten.
 +- de lichen zorgen voor de contaminatie. Ik heb de lichen zo goed mogelijk gewassen en de plukjes hout/aarde waar de lichen op groeide eraf gepulkt, maar dit is microskopisch werk en helemaal verdwenen zijn die substraatstrukjes natuurlijk niet.
 +- ook de Synechocystis kan gecontamineerd zijn. Ik heb die gekregen van Sandrine, maar ik heb hem verder niet steriel behandeld. Die Synechocystis culturen groeien al zo lang bij mij, en worden steeds (bij hun groei) terug aangelengd met BG11 maar niet in steriele omgeving …
